Pantothenic acid, also called vitamin B5 (a B vitamin), is a water-soluble vitamin. Pantothenic acid is an essential nutrient. Calcium Pantothenate or D-calcium pantothenate is the calcium salt of the water-soluble vitamin B5, ubiquitously found in plants and animal tissues with antioxidant property. Calcium Pantothenate is the calcium salt of pantothenic acid and is used in a combination with other vitamins to treat vitamin deficiency conditions.

Calcium Pantothenate is the calcium salt of the dextrorotatory isomer of pantothenic acid. It contains not less than 5.7 percent and not more than 6.0 percent of nitrogen (N), and not less than 8.2 percent and not more than 8.6 percent of calcium (Ca), both calculated on the dried basis.

Packaging and storage: Preserve in tight containers.
Identification:
A: Infrared Absorption.
B: A solution (1 in 20) responds to the tests for Calcium.
Specific rotation: between +25.0 and +27.5 .
Test solution: 50 mg per mL, in water.
Alkalinity: Dissolve 1.0 g in 15 mL of carbon dioxide-free water in a small flask. As soon as solution is complete, add 1.0 mL of 0.10 N hydrochloric acid, then add 0.05 mL of phenolphthalein, and mix: no pink color is produced within 5 seconds.
Loss on drying: Dry it at 105C for 3 hours: it loses not more than 5.0% of its weight.
Heavy metals: ---: the limit is 0.002%.
Nitrogen content: To pass the test.
Content of calcium: Dissolve about 800 mg of Calcium Pantothenate, accurately weighed, in 150 mL of water containing 2 mL of 3 N hydrochloric acid. Add 15 mL of 1 N sodium hydroxide and 300 mg of hydroxy naphthol blue, and titrate with 0.05 M edetate disodium until the solution is a distinct blue in color. Each mL of 0.05 M edetate disodium is equivalent to 2.004 mg of Ca.

DEFINITION
Calcium pantothenate contains not less than 98.0 per cent and not more than the equivalent of 101.0 per cent of calcium bis[3-[[(2R)-2,4-dihydroxy-3,3-dimethylbutanoyl]amino]propanoate], calculated with reference to the dried substance.


CHARACTERS
A white or almost white powder, slightly hygroscopic, freely soluble in water, slightly soluble in alcohol.


IDENTIFICATION
A. Specific optical rotation.
B. Examine the chromatograms.
C. To 1 mL of solution S (see Tests) add 1 mL of dilute sodium hydroxide solution and 0.1 mL of copper sulfate solution. A blue colour develops.
D. It gives reaction (a) of calcium.


TESTS
Solution S: Dissolve 2.50 g in carbon dioxide-free water and dilute to 50.0 mL with the same solvent.
Appearance of solution: Solution S is clear and colourless.
pH: The pH of solution S is 6.8 to 8.0.
Specific optical rotation: + 25.5 to + 27.5, determined on solution S and calculated with reference to the dried substance.
3-Aminopropionic acid: To pass the test.
Chlorides: 5 mL of solution S diluted to 15 mL with water complies with the limit test for chlorides (200 ppm).
Loss on drying: Not more than 3.0 per cent, determined on 1.000 g by drying in an oven at 105C.


ASSAY
Dissolve 0.180 g in 50 mL of anhydrous acetic acid. Titrate with 0.1 M perchloric acid determining the end-point potentiometrically.
1 mL of 0.1 M perchloric acid is equivalent to 23.83 mg of C18H32CaN2O10.

DESCRIPTION
Calcium Pantothenate occurs as a slightly hygroscopic, white powder. It is the calcium salt of the dextrorotatory isomer of pantothenic acid. It is stable in air. One gram dissolves in about 3 mL of water. It is soluble in glycerin, but is practically insoluble in alcohol, in chloroform, and in ether.
Function: Nutrient.


REQUIREMENTS
Identification:
A. A 1:20 aqueous solution gives positive tests for Calcium.
B. The infrared absorption spectrum of a potassium bromide dispersion of the sample, previously dried at 105C for 3 h, exhibits relative maxima at the same wavelengths as those of a similar preparation of USP Calcium Pantothenate.
C. Boil 50 mg of sample in 5 mL of 1 N sodium hydroxide for 1 min, cool, and add 5 mL of 1 N hydrochloric acid and 2 drops of ferric chloride. A strong yellow color appears.
Assay: Not less than 97.0% and not more than 103.0% of Dextrorotatory Calcium Pantothenate (C18H32CaN2O10) after drying.
Alkalinity: Passes test.
Alkaloids: Passes test.
Calcium Content: Not less than 8.2% and not more than 8.6% of calcium (Ca) after drying.
Lead: Not more than 2 mg/kg.
Loss on Drying: Not more than 5.0%.
Optical (Specific) Rotation 25C: Between +25.0° and +27.5° after drying.
